I have been trying to get myself to do a charity ride for the last 2 years or so but haven't been able to get motivated or fit one into my schedule during the summer.
This year a friend told me about the Tour de Cure put on by the American Diabetes Association and lucky for me its early in the year before my season really gets going. It's only 50 miles but that will be a good warm up and I am thinking about trying the ride on my fixed gear as an extra challenge. The ride is down in Falmouth on the Cape, I haven't spent much time down that way and touring around on my bike will be a good way to take in the sights.
oh and while your reading this...donate some money to the cause:

*UPDATE*: This ride is DUN. didnt end up doing it on my fixed gear, I wanted to get in more miles on my road bike to get ready for the B2B and I had done 60 miles the day before and 50 on the fixie after that would be rough. Weather was 'okay' at best, cold at times but at least the rain held off for the most part.
The route was really cool, got to see all the Cape Cod staples: ocean views, cranberry bogs, huge mansions. Ride was well supported with 3 rest stops fully stocked with food, water, gatorade, etc. Our team raised over $1,700 dollars for charity...we never got any numbers from ADA but I think we may have raised the most money out of all the teams.
I might post some pictures later...didnt bring the camera on the ride due to rain but there are some pre/post ride pics floating around.
